  • RRID:Addgene_24501

http://www.addgene.org/24501

Species: Mus musculus
Genetic Insert: Gq alpha
Vector Backbone Description: Backbone Size:4000; Vector Backbone:pcDNA1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: This is Gq alpha with the C-terminal amino acids changed from Gq alpha to Gi alpha residues (EYNLV to DCGLF). This construct allows many Gi-coupled receptors to stimulate phospholipase C (PLC). This is the most popular chimera, perhaps because it is easier to talk about coupling to Gi-coupled receptors with "qi5" rather than "qo5" or "qz5." Contains an internal hemaglutinin epitope tag (DVPDYA) that has no effect on receptor coupling. Please see supplemental document for G-protein chimera user manual.

  • RRID:Addgene_24500

http://www.addgene.org/24500

Species: Mus musculus
Genetic Insert: Gq alpha
Vector Backbone Description: Backbone Size:4000; Vector Backbone:pcDNA1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: This is Gq alpha with the C-terminal amino acids changed from Gq alpha to Go alpha residues (EYNLV to GCGLY). Works the same as qi5 but (for unknown reasons) has a slightly lower basal PLC activity. This can increase the signal-to-noise ratio. Contains an internal hemaglutinin epitope tag (DVPDYA) that has no effect on receptor coupling. Please see supplemental document for G-protein chimera user manual.

Proper citation: RRID:Addgene_24500 Copy

  • RRID:Addgene_24498

http://www.addgene.org/24498

Species: Mus musculus
Genetic Insert: Gq alpha
Vector Backbone Description: Backbone Size:4000; Vector Backbone:pcDNA1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: This is Gq alpha with the C-terminal amino acids changed from Gq alpha to Gs alpha residues (EYNLV to QYELL). This construct allows some Gs-coupled receptors to stimulate phospholipase C. Contains an internal hemaglutinin epitope tage (DVPDYA) that has no effect on receptor coupling. Please see supplemental document for G-protein chimera user manual.

Genetic Insert: RAG-1
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:4356; Vector Backbone:pCDM8; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: cDNA was synthesized by reverse transcription of total RNA obtained from cell line 22D6, an Abelson murine leukemia virus transformed pre-B cell line. RAG-1 cDNA was then amplified by PCR, using specific primers designed also to add restriction enzyme recognition sites adjacent to the largest open reading frame for subsequent cloning. Species: Mus musculus
Genetic Insert: CHOP10
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:4000; Vector Backbone:pcDNA1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: From depositing lab: "9E10 (Myc-epitope) tagged mouse CHOP coding region only. The insert was joined to the epitope tag using patch-PCR and incorporating unique BamH1 and Xho1 sites for ligation into pcDNA1. Stocks are provided as MC1060 (or MC1061, we are not sure) host strain transformed with the plasmid. Note that pCDNA1 plasmids must be propagated on combined amp and tet selection. I would recommedn that you consider transferring the insert into a better host strain such as pcDNA3"

  • RRID:Addgene_24499

http://www.addgene.org/24499

Species: Rattus norvegicus
Genetic Insert: Gs alpha
Vector Backbone Description: Backbone Size:4000; Vector Backbone:pcDNA1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin and Tetracycline
References:
Comments: This is Gs alpha with the C-terminal amino acids changed from Gs alpha to Gq alpha residues (QYELL to EYNLV). This construct allows some Gq-coupled receptors to stimulate Adenylate cyclase. There isn't much experience with this chimera at the moment. It may be useful for people who find the AC stimulation is better readout of receptor activation than PLC stimulation. Contains an internal hemaglutinin epitope tage (DVPDYA) that has no effect on receptor coupling. Please see supplemental document for G-protein chimera user manual.
